Indication
- Treatment of panic disorder
- Treatment of panic disorder with or without agoraphobia

Dosage & Administration
- Initial adult dose not to exceed 1.5 mg/day
- Increased dosage in increments of 0.5 to 1 mg every 3 days
- Maximum recommended daily dose is 20 mg
- Initial adult dose for panic disorder is 0.25 mg
- Initial pediatric dose for infants and children is between 0.01 and 0.03 mg/kg/day
Interaction
- No alteration in pharmacokinetics of phenytoin
- No alteration in pharmacokinetics of carbamazepine
- No alteration in pharmacokinetics of phenobarbital
- Effect on metabolism of other drugs not investigated
Contraindications
- History of hypersensitivity to benzodiazepines
- Clinical or biochemical evidence of significant liver disease
- Contradicted in acute narrow angle glaucoma
Side Effects
- Frequently occurring side effects referable to CNS depression
- Drowsiness in approximately 50% of patients
- Ataxia in approximately 30% of patients
- Behavior problems noted in approximately 25% of patients
Pregnancy & Lactation
- Potential to produce congenital malformations
- Evidence that anticonvulsant drugs act as teratogens
- Compelling indication for administration during pregnancy
Precautions & Warnings
- May increase the incidence of generalized tonic-clonic seizures
- Concomitant use with valproic acid may produce absence status
Use in Special Populations
- Increased production of saliva and bronchial secretion in infants and small children
- Benzodiazepine pharmacologic effects appear to be greater in elderly patients
- Renal impairment does not affect the pharmacokinetics
- Influence of hepatic impairment on pharmacokinetics not fully investigated
Overdose Effects
- Causes drowsiness, ataxia, dysarthria, and nystagmus
- May lead to areflexia, apnoea, hypotension, and coma
- Benzodiazepine respiratory depressant effects more serious in patients with respiratory disease
Therapeutic Class
- Adjunct anti-epileptic drugs
- Benzodiazepine hypnotics
Reconstitution
- Contents of the vial must be diluted with 1 ml of water for injection prior to administration
- Injection solution should be prepared immediately before use
- IV injection should be administered slowly with continuous monitoring
