Description

  • Pyrrolo-pyrrole group nonsteroidal anti-inflammatory drug (NSAID)
  • Chemical compound structure: 5 benzoyle-2,3-dihydro-1H-pyrroligine-1-carboxylic acid, compound with 2-amino-2-(hydroxymethyl)-1,3-propanediol
  • Inhibits prostaglandin synthesis
  • Peripherally acting analgesic
  • Biological activity associated with S-form
  • Linear pharmacokinetic property
  • Highly protein bound
  • Largely metabolized in the liver
  • Metabolites and unchanged drugs excreted in urine

Pharmacology

  • Potent analgesic
  • Non-steroidal anti-inflammatory drug (NSAID)
  • Inhibits cyclooxygenase enzyme system
  • Minimal anti-inflammatory effect at analgesic dose

Dosage & Administration

  • Recommended tablet dose: 10 mg every 4-6 hours for short-term use (up to 7 days)
  • Injection: Single or multiple doses for moderately severe acute pain management, with specific dosing guidelines based on age, weight, and condition
  • Eye drops: Recommended use of 1 drop in each eye 4 times daily

Interaction

  • Increased side effects with other NSAIDs or aspirin
  • Enhanced anti-coagulant effect with anti-coagulants
  • Reduced anti-hypertensive effect with beta blockers
  • Increased risk of renal impairment with ACE inhibitors
  • Enhanced toxicity of methotrexate

Contraindications

  • Hypersensitivity to Ketorolac or other NSAIDs
  • Not for use in children under 16 years old
  • Contraindicated as prophylactic analgesic before surgery

  • Avoid administering while wearing contact lenses
