Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). It binds with high affinity to the alpha2-delta site in central nervous system tissues, which may be involved in its anti-nociceptive and antiseizure effects.
Dosage
Recommended doses for various conditions including neuropathic pain, postherpetic neuralgia, fibromyalgia, and spinal cord injury.
Administration
- Route of administration: Oral
- Administration instructions: Should be swallowed whole and should not be split, crushed or chewed. Pregabalin CR tablet should be administered after an evening meal.
Interaction
- Drug interaction with medication: Unlikely to be involved in significant pharmacokinetic drug interactions
- Drug interaction with food and others: Not applicable
Contraindications
Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components

Pregnancy & Lactation
- Pregnancy: No adequate and well-controlled studies with pregabalin in pregnant women. Pregnant women should be advised of the potential risk to a fetus
- Lactation: Small amounts of pregabalin have been detected in the milk of lactating women. Breastfeeding is not recommended
Precautions & Warnings
- Angioedema can occur and may be associated with life-threatening respiratory compromise
- Hypersensitivity reactions
- Increased risk of suicidal thoughts or behavior
- Respiratory depression may occur when used with concomitant CNS depressants
- Impairment of ability to drive or operate machinery
- Caution when coadministering with thiazolidinedione antidiabetic agents
Use in Special Populations
- Children and adolescents: Safety and effectiveness not established for various conditions
Overdose Effects
Signs and symptoms include reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, and heart block. No specific antidote.
