Description
Charm is a selective, reversible inhibitor of cyclic guanosine monophosphate (cGMP)-specific phosphodiesterase type 5 (PDE5)
Mechanism of Action
Inhibition of PDE5 by Charm produces increased levels of cGMP in the corpus cavernosum, resulting in smooth muscle relaxation and inflow of blood into the penile tissues, thereby producing an erection
Pharmacokinetics
Mean absolute bioavailability of 41%, mostly hepatic metabolism via cytochrome P450 3A4, terminal half lives of about 4 hours
Dosage & Administration
- Recommended dose of 50 mg, may be increased to a maximum dose of 100 mg
- The maximum recommended dosing frequency is once per day
Drug Interactions
Metabolism affected by CYP3A4 inhibitors, interaction with nitrates can lead to potentiation of hypotensive effects
Contraindications
- Hypersensitivity to any component of the medication
- Use with organic nitrates

- Assessment of potential underlying causes of erectile dysfunction
- Caution with concurrent use of alpha-blockers
- Potential for cardiac risk in patients with preexisting cardiovascular disease
- Sensitivity to actions of vasodilators
- Use with caution in various patient groups
