Indications
- Anaplastic lymphoma kinase (ALK)-positive metastatic Non-small cell lung cancer (NSCLC) treatment
- Progressed on or intolerant to Crizotinib
Pharmacology
- Tyrosine kinase inhibitor
- Inhibits ALK, ROS1, IGF-1R, and FLT-3
- Inhibits autophosphorylation of ALK and downstream signaling proteins
- Metabolized by CYP2C8 and CYP3A4
- Excretion in feces and urine
Dosage & Administration
- Recommended initial dose: 90 mg orally once daily for the first 7 days
- If tolerated, increase to 180 mg orally once daily
- Administer until disease progression or unacceptable toxicity
- May be taken with or without food
- Tablets should be swallowed whole
Interaction
- Strong CYP3A Inhibitors increase plasma concentrations
- Strong CYP3A Inducers decrease plasma concentrations
- Biganib may alter plasma concentrations of CYP3A substrates
Contraindications
- Known hypersensitivity to Brigatinib or any other components

- Creatine Phosphokinase (CPK) Elevation
- Pancreatic Enzyme Elevation
- Hyperglycemia
Pregnancy & Lactation
- No clinical data on use in pregnant women
- Adverse reactions in breastfed infants, lactating women advised not to breastfeed during treatment
- Females of reproductive potential advised to use effective non-hormonal contraception
- Males with female partners advised to use effective contraception
- May cause reduced fertility in males

- Severe pulmonary adverse reactions (ILD/pneumonitis)
- Hypertension monitoring and control
- Bradycardia monitoring
- Visual symptoms evaluation
- CPK and pancreatic enzyme elevation monitoring
- Hyperglycemia monitoring and management
