Indications
- Treatment of adult patients with ALK-positive advanced non-small cell lung cancer
- Treatment after progression on other ALK inhibitor therapies
- Monotherapy for adult patients
Pharmacology
- Kinase inhibitor targeting ALK and ROS1
- Inhibitor of TYK1, FER, FPS, TRK family, FAK, FAK2, and ACK
- Activity against multiple mutant forms of the ALK enzyme
- In vitro and in vivo antitumor activity
Dosage & Administration
- Recommended dose of 100mg orally once daily
- Dosing interruption or dose reduction may be required based on safety and tolerability
Interaction
- CYP3A4/5 time-dependent inhibitor and inducer
- Potential to inhibit CYP2C9 and induce CYP2B6
- Inhibition of drug transporters such as P-gp and BCRP
Contraindications
- Hypersensitivity to Lorlatinib
- Concomitant use of strong CYP3A4/5 inducers

Use in Special Populations
