Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). It does not bind directly to GABA receptors, but binds with high affinity to the alpha2-delta site in central nervous system tissues, involved in anti-nociceptive and antiseizure effects.
Maximum Recommended Dose
330 mg/day for adults with neuropathic pain associated with diabetic peripheral neuropathy
Recommended Dosage for Postherpetic Neuralgia in Adults
75 to 150 mg two times a day or 50 to 100 mg three times a day (150 to 300 mg/day)
Recommended Dosage for Fibromyalgia in Adults
300 to 450 mg/day
Recommended Dosage for Neuropathic Pain Associated with Spinal Cord Injury in Adults
150 to 600 mg/day
Route of Administration
Oral, can be taken with or without food
Pregnancy & Lactation
Pregabalin is not recommended during pregnancy and breastfeeding due to potential risk to the fetus and presence in the milk of lactating women
Precautions
- Angioedema may occur and require immediate discontinuation
- Increased risk of suicidal thoughts or behavior
- Respiratory depression may occur when used with concomitant CNS depressants
- May cause dizziness and somnolence, impairing the ability to drive or operate machinery
- Caution should be exercised when coadministering with antidiabetic agents
- Discontinuation should be gradual over a minimum of 1 week
Use in Special Populations
Safety and effectiveness in pediatric patients have not been established for certain uses
