Indications
- Chronic constipation treatment
- Adults only

Pharmacology
- Selective stimulator of 5-HT4 receptors
- No interaction with hERG channel
- Stimulation of propulsive motility
Dosage & Administration
- Adult dosage: 2 mg once daily
- Older people dosage: Start with 1 mg once daily
- Children: Not for use in children and adolescents
Interaction
- Low interaction potential
- No expected effect on CYP-mediated metabolism
- Potential interaction with potent P-gp inhibitors
Contraindications
- Hypersensitivity to active substance or excipients
- Renal impairment requiring dialysis
Side Effects
- Frequently reported: headache, gastrointestinal symptoms
- Adverse reactions occur predominantly at the start of therapy
Pregnancy & Lactation
- Not recommended during pregnancy
- Effective contraception recommended for women of childbearing potential
- Not recommended during breastfeeding
Precautions & Warnings
- Renal excretion as main elimination route
- Caution in severe hepatic impairment
- Efficacy of oral contraceptives may be reduced in severe diarrhoea
- Tablets contain lactose, caution in patients with galactose intolerance
Use in Special Populations
- Renal impairment dose: 1 mg once daily for GFR <30 ml/min/1.73 m2
- Hepatic impairment dose: Start with 1 mg once daily for Child-Pugh class C
Overdose Effects
- Exaggeration of known pharmacodynamic effects
- Symptomatic treatment for overdose
- Correction of electrolyte disturbances may be required
