  • Regression of ascites secondary to carcinoma
  • Reduction of bleeding during surgical interventions

Pharmacology

  • Strong antifibrinolytic action
  • Inhibition of plasminogen activation
  • Low acute toxicity
  • Well absorbed by oral route
  • Excretion by renal route
  • No interference with clotting processes
  • No tendency to thrombophilia

Dosage & Administration

  • Recommended dose for adults: 500-1000 mg 3 times daily
  • Mean recommended daily doses for prophylaxis: 0.5-1 gm orally, 500 mg by parenteral route
  • Oral dose increases to 1-3 gm for therapy of hemorrhagic manifestations
  • Orally administered daily in divided doses for children
  • No reduction in dosage for elderly patients

Interaction

  • Incompatible with solutions containing penicillins
  • Antagonizes the antifibrinolytic action of thrombolytic drugs
  • Increased potential for thrombus formation with concomitant estrogen-containing drugs
  • Avoid direct admixture with whole blood during transfusion

Contraindications

  • Individual hypersensitivity
  • Thromboembolic disease
  • Arterial and venous thrombosis
  • Endocavitary hemorrhages
  • Serious kidney failure

Pregnancy & Lactation

  • Avoid during known and presumed pregnancy
  • Passes into breast milk at low concentration

Precautions & Warnings

  • Use in cases of hyperfibrinolysis
  • Prophylactic treatment 24 hours before operation
  • Prolonged therapy for at least 24 hours after manifestations have disappeared
  • Care in cases of hematuria
  • Avoid in serious renal insufficiency or anuric syndromes
  • Use with caution in less serious renal dysfunctions
  • Particular care in cardiopathic and hepatopathic subjects
