Pharmacology
- Strong antifibrinolytic action
- Inhibition of plasminogen activation
- Low acute toxicity
- Well absorbed by oral route
- Excretion by renal route
- No interference with clotting processes
- No tendency to thrombophilia
Dosage & Administration
- Recommended dose for adults: 500-1000 mg 3 times daily
- Mean recommended daily doses for prophylaxis: 0.5-1 gm orally, 500 mg by parenteral route
- Oral dose increases to 1-3 gm for therapy of hemorrhagic manifestations
- Orally administered daily in divided doses for children
- No reduction in dosage for elderly patients
Interaction
- Incompatible with solutions containing penicillins
- Antagonizes the antifibrinolytic action of thrombolytic drugs
- Increased potential for thrombus formation with concomitant estrogen-containing drugs
- Avoid direct admixture with whole blood during transfusion

- Individual hypersensitivity
- Thromboembolic disease
- Arterial and venous thrombosis
- Endocavitary hemorrhages
- Serious kidney failure

Pregnancy & Lactation
- Avoid during known and presumed pregnancy
- Passes into breast milk at low concentration
Precautions & Warnings
- Use in cases of hyperfibrinolysis
- Prophylactic treatment 24 hours before operation
- Prolonged therapy for at least 24 hours after manifestations have disappeared
- Care in cases of hematuria
- Avoid in serious renal insufficiency or anuric syndromes
- Use with caution in less serious renal dysfunctions
- Particular care in cardiopathic and hepatopathic subjects
