Indications
- Treatment of thrombocytopenia in patients with persistent or chronic immune thrombocytopenia
- Treatment of thrombocytopenia in patients with hepatitis C infection
- Treatment of severe aplastic anemia
Pharmacology
- Small molecule thrombopoietin (TPO) receptor agonist
- Interacts with the transmembrane domain of the human TPO-receptor
- Induces proliferation and differentiation from bone marrow progenitor cells
Dosage & Administration
- Route of administration: oral
- Can be taken with or without a meal low in calcium
- Specific dosages for different conditions and patient groups
Interaction
- Drug interaction with medication: take at least 2 hours before or 4 hours after certain medications or products containing polyvalent cations
- Drug interaction with food: take at least 2 hours before or 4 hours after calcium-rich foods
Contraindications
- Contraindicated in patients with known hypersensitivity to eltrombopag or any components
Side Effects
- Common side effects: anemia, nausea, pyrexia, ALT increased, cough, fatigue, headache, diarrhea
Pregnancy & Lactation
- Insufficient data available for assessing risks of major birth defects, miscarriage, or adverse maternal or fetal outcomes
- Not recommended during breastfeeding due to potential for serious adverse reactions in the child
Precautions & Warnings
- Increased risk of hepatic decompensation in patients with chronic hepatitis C receiving R-Pag in combination with interferon and ribavirin
- Increased risk of death and progression of myelodysplastic syndromes to acute myeloid leukemia
- Reports of portal vein thrombosis in patients with chronic liver disease receiving eltrombopag
Use in Special Populations
- Safety and efficacy in pediatric patients below the age of 1 year with ITP have not been established
- Safety and efficacy in pediatric patients with thrombocytopenia associated with chronic hepatitis C and refractory severe aplastic anemia have not been established
Overdose Effects
- Platelet counts may increase excessively and result in thrombotic or thromboembolic complications
- Consider oral administration of metal cation containing preparation in case of overdose to limit absorption
