Indications
- Spasticity resulting from multiple sclerosis
- Flexor spasms and concomitant pain
- Clonus and muscular rigidity
- Skeletal muscle spasm resulting from rheumatic disorders
- Spinal cord injuries and other spinal cord diseases
- Cerebrovascular accidents or neoplastic or degenerative brain disease
Pharmacology
Inhibition of monosynaptic and polysynaptic reflexes by stimulating GABA receptors, CNS depression, antinociceptive effect
Dosage & Administration
- Adult & children over 10 years: 5 mg 3 times daily, preferably with or after food, gradually increased; max. 100 mg daily
- Children <10 years: Treatment usually started with 2.5 mg (2.5 ml) given 4 times daily then raised according to requirement
Interactions
- Increased sedation with agents acting on the central nervous system
- Increase in fall in blood pressure when combined with antihypertensives
- Potentiated pharmacological effects when combined with tricyclic antidepressants
- Reports of mental confusion, hallucinations, headaches, nausea and agitation with Parkinson's treatment
- Increased CNS depressant effects with MAO inhibitors
- Possible synergistic effect with magnesium sulphate or other neuromuscular blocking agents

Pregnancy & Lactation
- Category: B3
- Use during pregnancy: Safe use not established, crosses placental barrier
- Use during lactation: Excreted in breast milk in small quantities
Precautions & Warnings
- Lower doses for impaired renal function
- Cautious treatment for patients with psychotic disorders, schizophrenia, depressive or manic disorders
- Possible lowering of the convulsion threshold in patients with epilepsy
- Use with caution in patients with peptic ulcers, cerebrovascular diseases, or hepatic, renal or respiratory failure
- Careful monitoring of respiratory and cardiovascular function
- Potential improvement of neurogenic disturbances affecting emptying of the bladder
- Poor tolerance by stroke patients
- Regular laboratory tests for patients with hepatic diseases or diabetes mellitus
Overdose Effects
Gastric lavage important in case of severe overdose
