Pharmacology

  • Tranexamic acid
  • Antifibrinolytic action
  • Inhibition of plasminogen activation
  • Low acute and chronic toxicity
  • Well absorbed by oral route
  • Excretion by renal route
  • No interference with clotting processes

Dosage & Administration

  • Usual adult dose: 500-1000 mg 3 times daily
  • Mean recommended daily doses for prophylaxis: 0.5-1 gm orally, 500 mg parenterally
  • Increased oral dose for therapy of hemorrhagic manifestations
  • Dosage for children based on body weight

Interaction

  • Incompatibility with penicillins
  • Antagonism with thrombolytic drugs
  • Increased potential for thrombus formation with estrogen-containing drugs
  • Avoid direct admixture with whole blood during transfusion

Contraindications

  • Known hypersensitivity to the product
  • Thromboembolic disease
  • Arterial and venous thrombosis
  • Endocavitary hemorrhages
  • Serious kidney failure

Pregnancy & Lactation

  • Avoid use during known and presumed pregnancy
  • Passage into breast milk at low concentration

Precautions & Warnings

  • Use in hyperfibrinolysis cases
  • Initiate prophylactic treatment before operation
  • Prolong therapy of hemorrhages
  • Adjust doses in hematuria
  • Avoid use in serious renal insufficiency
  • Exercise caution in renal and hepatic dysfunctions
