Dosage Administration
- Oral: Swallow whole, taken one hour before a meal
- Oral Suspension: Mixed with water, swallowed or administered through nasogastric or gastric tube
- IV Injection: Intravenous administration over at least 3 minutes

- Symptomatic GERD: 20 mg once daily for 4 weeks
- Helicobacter Pylori eradication: Triple Therapy for 10 days
- Zollinger-Ellison Syndrome: 20-80 mg once daily
- Acid-related Dyspepsia: 20-40 mg once daily for 2-4 weeks
- Duodenal Ulcer: 20 mg once daily for 2-4 weeks
- Gastric Ulcer: 20-40 mg once daily for 4-8 weeks
- IV Injection for Adult and Pediatric Patients
Pharmacology
- Mechanism of Action: Specific inhibition of H+/K+-ATPase in the gastric parietal cell
- Absorption: Enteric-coated pellet formulation, peak plasma levels at approximately 1.5 hours after oral administration
- Distribution: 97% bound to plasma proteins
- Metabolism: Extensively metabolized by the CYP enzyme system
- Excretion: Plasma elimination half-life of approximately 1-1.5 hours, less than 1% excreted unchanged in urine
Interaction
- Metabolized by CYP2C19 and CYP3A4
- No clinically significant interactions with certain drugs identified
- Potential interference with CYP2C19 and gastric pH-dependent drug absorption
- Increased plasma levels when co-administered with certain drugs
