Indications

Pharmacology

Contraindications

  • Hypersensitivity to trimethoprim or sulphonamides
  • Megaloblastic anaemia due to folate deficiency
  • Liver parenchymal damage
  • Blood dyscrasia
  • Severe renal insufficiency
  • Glucose 6-phosphate dehydrogenase deficiency

Side Effects

  • Rare side effects such as crystalluria, haemolysis, thrombocytopenia
  • Less serious side effects like malaise, headache, nausea, vomiting
  • Transient nature of side effects

Pregnancy & Lactation

  • Risk of passing sulphonamides to the placenta
  • Excretion of sulphonamides in breast milk
  • Potential for causing kernicterus

Precautions & Warnings

  • Risk of macrocytic anaemia with prolonged treatment
  • Interference in the conversion of Folic acid into Folinic acid
  • Reversible with Folinic acid supplementation
  • Caution in diabetic patients receiving sulphonylurea drugs
