Description
- Domp 5 mg/5 ml Suspension is a dopamine antagonist medication used to treat nausea and vomiting
- It helps relieve dyspeptic symptoms, acute nausea and vomiting, Parkinson's disease related symptoms, and speeding barium transit in follow-through radiological studies
- It restores normal motility and tone of the upper gastrointestinal tract, facilitates gastric emptying, enhances antral and duodenal peristalsis, regulates contraction of the pylorus, increases esophageal peristalsis and lower esophageal sphincter pressure, and prevents regurgitation of gastric content
- It interacts with anticholinergic drugs, antacids, antisecretory drugs, CYP3A4 inhibitors, neuroleptics, and dopaminergic agonists
DosageAndAdministration
- Should be taken 15-30 minutes before meals and, if necessary, before retiring
- Recommended adult dose is 10-20 mg (1-2 tablets or 10-20 ml suspension), every 6-8 hours daily
- Maximum dose is 80 mg daily
- In acute nausea and vomiting, adult dose is 20 mg (2 tablets or 20 ml suspension), every 6-8 hours daily
- Rectal administration: adult dose is 30-60 mg every 4-8 hours
Pharmacology
- Dopamine antagonist that blocks dopamine receptors located in the Chemoreceptor Trigger Zone (CTZ) and stomach
- Restores normal motility and tone of the upper gastrointestinal tract, facilitates gastric emptying, enhances antral and duodenal peristalsis, regulates contraction of the pylorus, and increases esophageal peristalsis and lower esophageal sphincter pressure
Interaction
- Antagonises the antidyspeptic effect of domperidone when used with anticholinergic drugs
- Antacids and antisecretory drugs lower its oral bioavailability
- Concomitant use of CYP3A4 inhibitors may result in increased plasma levels of domperidone
Contraindications
- Known hypersensitivity to domperidone, neonates, gastrointestinal hemorrhage, mechanical obstruction or perforation, and prolactin releasing pituitary tumor
SideEffects
- Transient intestinal cramps, extrapyramidal phenomena (rare), increase in plasma prolactin level, rare allergic reactions such as rash and urticaria
PregnancyAndLactation
- Should only be used during the first trimester of pregnancy if justified by therapeutic benefit
- Excreted in breast milk; not recommended for nursing mothers
PrecautionsAndWarnings
- Absolute caution in the case of children due to increased risk of extra-pyramidal reactions
- Caution in patients with hepatic impairment
