Ezetim 10 mg (Tablet)

Indications

• Primary hypercholesterolemia
• Prevention of cardiovascular events
• Homozygous familial hypercholesterolaemia (HoFH)
• Homozygous sitosterolemia (Phytosterolemia)

Pharmacology

• Localizes at the brush border of the small intestine
• Inhibits absorption of cholesterol via NPC1L1 sterol transporter
• Decreases delivery of cholesterol to the liver
• Reduces hepatic cholesterol stores
• Increases clearance of cholesterol from the blood

Dosage & Administration

• Recommended dose: 10mg once daily
• Can be administered with or without food

Interaction

• Fibrates may increase cholesterol excretion into the bile
• Coadministration with fibrates not recommended

Contraindications

• Hypersensitivity to any component
• Active liver disease
• Unexplained persistent elevations in serum transaminases

Side Effects

• Generally well tolerated
• Overall incidence of adverse events similar to placebo
• Discontinuation rate due to adverse events similar to placebo

Pregnancy & Lactation

• Limited studies in pregnant women
• Use only if potential benefit justifies risk to the fetus

Precautions & Warnings

• Exclude or treat secondary causes of dyslipidaemia
• Renal and hepatic impairment
• Not recommended for use in children < 10 years

Use in Special Populations

• Pediatric Use
• 10 to 17 years: No dosage adjustment required
• Limited clinical experience in pediatric and adolescent patients

Overdose Effects

• No reported cases of overdosage
• Administration of 50 mg/day generally well tolerated
• Symptomatic and supportive measures in case of overdose
