Ezetrol 10 mg (Tablet)

Indications

• Adjunctive therapy for primary hypercholesterolemia
• Prevention of cardiovascular events in patients with coronary heart disease
• Treatment for homozygous familial hypercholesterolaemia
• Adjunctive therapy for homozygous sitosterolemia

Pharmacology

• Localizes at the brush border of the small intestine
• Inhibits absorption of cholesterol via sterol transporter NPC1L1

Dosage & Administration

• Recommended dose of 10mg once daily
• Can be taken with or without food

Interaction

• Potential interaction with fibrates increasing cholesterol excretion into bile

Contraindications

• Hypersensitivity to any component of the medication
• Contraindicated in patients with active liver disease or unexplained persistent elevations in serum transaminases

Side Effects

• Generally well tolerated in clinical studies
• Similar incidence of adverse events reported with placebo
• Similar discontinuation rate due to adverse events as placebo

Pregnancy & Lactation

• No adequate and well-controlled studies in pregnant women
• Should be used during pregnancy only if potential benefit justifies the risk to the fetus

Precautions & Warnings

• Exclude or treat secondary causes of dyslipidaemia prior to initiating therapy
• Consider renal and hepatic impairment
• Caution advised during pregnancy and lactation

Use in Special Populations

• Limited clinical experience in pediatric and adolescent patients
• No dosage adjustment required for patients aged 10 to 17 years
• Not recommended for children below age 10 due to insufficient data on safety and efficacy

Overdose Effects

• No reported cases of overdosage
• 50 mg/day dose generally well tolerated in clinical trial
