Indications
- Symptomatic treatment of chronic constipation in adults
Pharmacology
- Selective stimulator of the 5-HT4 receptors
- Acts primarily on smooth muscle and myenteric plexus
- Stimulates motility by releasing acetylcholine
- Propels luminal content through the GI tract
Dosage & Administration
- Adult dosage: 2 mg once daily with or without food
- Start with 1 mg for older people
- Not recommended for children under 18 years
Interaction
- Low interaction potential with CYP-mediated metabolism
- May have minor interactions with potent inhibitors of P-gp
- No clinically relevant effects on certain drugs' pharmacokinetics
Contraindications
- Hypersensitivity to active substance or excipients
- Renal impairment requiring dialysis
Side Effects
- Frequently reported adverse reactions including headache and gastrointestinal symptoms like abdominal pain, nausea, and diarrhea
- Majority of adverse events are mild to moderate in intensity

- Reduced efficacy of oral contraceptives in case of severe diarrhea
- Not suitable for patients with rare hereditary problems of galactose intolerance, Lapp lactase deficiency or glucose-galactose malabsorption
