Indications
- Symptomatic treatment of chronic constipation in adults when laxatives fail to provide relief
Pharmacology
- Selective stimulator of 5-HT4 receptors
- Acts on gastrointestinal smooth muscle cells and myenteric plexus
- Stimulates motility through release of acetylcholine
Dosage & Administration
- Adult dose: 2mg once daily with or without food
- Elderly dose: Start with 1mg daily, increase to 2mg if needed
- Not recommended for children under 18
Interaction
- Low interaction potential with CYP-mediated metabolism
- Weak substrate for P-glycoprotein
- Slight increase in systemic exposure with potent inhibitors of P-gp
Contraindications
- Hypersensitivity to active substance or excipients
- Renal impairment requiring dialysis
Side Effects
- Most frequent: headache, abdominal pain, nausea, diarrhea
- Adverse reactions occur at the start of therapy and usually disappear within a few days
Pregnancy & Lactation
- Not recommended during pregnancy
- Effective contraception should be used
- Not recommended during breastfeeding due to absence of human data
Precautions & Warnings
- Dose adjustment for severe renal impairment
- Caution with severe hepatic impairment
- Potential reduction in efficacy of oral contraceptives in case of severe diarrhea
- Contains lactose - caution in patients with rare hereditary galactose intolerance
