Indications
- Symptomatic treatment of chronic constipation in adults
Pharmacology
- Stimulates motility by interacting with 5-HT4 receptors in the GI tract
- Acts as a selective stimulator of the 5-HT4 receptors
- Produces the release of acetylcholine in the GI tract
Dosage & Administration
- 2 mg once daily for adults
- Start with 1 mg once daily for older people
- Not recommended for children and adolescents younger than 18 years
Interaction
- Low interaction potential with therapeutic concentrations
- Not an inhibitor of P-gp at clinically relevant concentrations
- No clinically relevant effects on the pharmacokinetics of certain medications
Contraindications
- Hypersensitivity to the active substance or excipients
- Renal impairment requiring dialysis
Side Effects
- Headache
- Gastrointestinal symptoms (abdominal pain, nausea, diarrhea)
- Adverse reactions occur predominantly at the start of therapy
Pregnancy & Lactation
- Not recommended during pregnancy
- Women of childbearing potential should use effective contraception during treatment
- Not recommended during breastfeeding based on animal studies
Precautions & Warnings
- Dose adjustment required in severe renal impairment
- Caution in prescribing to patients with severe hepatic impairment
- Possible reduction in efficacy of oral contraceptives with severe diarrhea
Use in Special Populations

Overdose Effects
- Symptoms include headache, nausea, and diarrhea
- Specific treatment not available for overdose
