Indications

  • Symptomatic treatment of chronic constipation in adults
  • Laxatives replacement in adults

Pharmacology

  • Selective stimulator of 5-HT4 receptors
  • Stimulates propulsive motility in the colon
  • Release of acetylcholine in the GI tract
  • No interaction with hERG channel or 5-HT1 receptors

Dosage & Administration

  • Adult dosage: 2 mg once daily with or without food
  • Start with 1 mg once daily for older people
  • Not recommended for children and adolescents under 18 years

Interaction

  • Low interaction potential with CYP-mediated metabolism
  • Weak substrate for P-glycoprotein (P-gp)
  • Potential interaction with potent inhibitors of P-gp such as verapamil and cyclosporine A

Contraindications

  • Hypersensitivity to the active substance or excipients
  • Renal impairment requiring dialysis

Side Effects

  • Frequently reported adverse reactions: headache, abdominal pain, nausea, diarrhea
  • Adverse reactions occur predominantly at the start of therapy

Pregnancy & Lactation

  • Not recommended during pregnancy
  • Use effective contraception during treatment
  • Not recommended during breastfeeding due to lack of human data

Precautions & Warnings

  • Renal excretion is the main route of elimination
  • Caution in patients with severe hepatic impairment
  • Possible reduced efficacy of oral contraceptives in case of severe diarrhea
  • Contains lactose, not recommended for patients with galactose intolerance
