Indications
- spasticity resulting from multiple sclerosis
- flexor spasms and concomitant pain, clonus and muscular rigidity
- skeletal muscle spasm resulting from rheumatic disorders
- spinal cord injuries and other spinal cord diseases
- cerebrovascular accidents or neoplastic or degenerative brain disease
Pharmacology
- inhibits both monosynaptic and polysynaptic reflexes
- stimulates the GABA B receptors
- inhibits the release of glutamate and aspartate
- may produce CNS depression
- exerts an antinociceptive effect
Dosage & Administration
- 5 mg 3 times daily for adults & children over 10 years
- gradually increased dosage
- max. 100 mg daily for adults & children over 10 years
- dosage varies for children under 10 years based on age
- preferably taken with or after food
Interaction
- Increased sedation with agents acting on the central nervous system, alcohol or synthetic opiates
- Potentiation and increased fall in blood pressure when combined with antihypertensives
- Pharmacological effects potentiation when combined with tricyclic antidepressants
- Reports of mental confusion, hallucinations, headaches, nausea and agitation with concurrent use with levodopa and carbidopa
- Increased CNS depressant effects with concurrent use of MAO inhibitors
- Potential synergistic effect with magnesium sulphate or other neuromuscular blocking agents
Contraindications
- Hypersensitivity to any component of the product

- Various adverse reactions associated with the central nervous system, cardiovascular system, gastrointestinal system, genitourinary system, and others
Pregnancy & Lactation
- Pregnancy category B3
- Crosses the placental barrier
- Should only be administered to pregnant women when potential benefits outweigh the possible hazards
- Excreted in breast milk in small quantities with no expected undesirable effects on the infant
Precautions & Warnings
- Lower doses for patients with impaired renal function or undergoing chronic hemodialysis
- Cautious treatment for patients with psychotic disorders, schizophrenia, depressive or manic disorders
- Possible lowering of the convulsion threshold in patients with epilepsy and muscle spasticity
- Use with caution in patients with peptic ulcers, cerebrovascular diseases, or hepatic, renal or respiratory failure
- Essential monitoring of respiratory and cardiovascular function, especially in patients with cardiopulmonary disease and respiratory muscle weakness
- Possibility of acute retention of urine in patients with preexisting sphincter hypertonia
- Limited benefit and poor tolerance in stroke patients
Overdose Effects
- Importance of gastric lavage in case of severe overdose
