Indications
- Treatment of ALK-positive metastatic Non-small cell lung cancer (NSCLC)
Pharmacology
- Inhibition of ALK, ROS1, IGF-1R, FLT-3, EGFR, EML4-ALK, and NPM ALK fusion proteins
- Autophosphorylation inhibition of ALK
- Metabolized by CYP2C8 and CYP3A4
- Excretion in feces and urine
Dosage & Administration
- Initial dose of 90 mg orally once daily for 7 days
- Increased to 180 mg orally once daily if tolerated
- Administered with or without food
- Tablets to be swallowed whole
Interaction
- Avoid concomitant use with strong CYP3A inhibitors
- Avoid concomitant use with strong CYP3A inducers
- May alter plasma concentrations of CYP3A substrates
Contraindications
- Known hypersensitivity to Brigatinib

- Visual Disturbance
- Creatine Phosphokinase (CPK) Elevation
- Pancreatic Enzyme Elevation
- Hyperglycemia

- No clinical data on use in pregnant women
- Adverse reactions in breastfed infants
- Effective non-hormonal contraception advised
