Indications
- Symptomatic relief of Osteoarthritis (OA)
- Symptomatic relief of Rheumatoid arthritis (RA)
- Symptomatic relief of Ankylosing spondylitis
- Symptomatic relief of acute gouty arthritis
- Short-term treatment of moderate pain associated with dental surgery
Pharmacology
- Potent, orally active cyclooxygenase-2 (COX-2) specific inhibitor
- Decreases clinical signs and symptoms with decreased potential for GI toxicity and effects on platelet aggregation
- Produces dose-dependent inhibition of COX-2 without inhibition of COX-1 at doses up to 150 mg daily
Dosage & Administration
- Recommended dose for Osteoarthritis: 30 mg once daily, increased dose of 60 mg once daily in some patients
- Recommended dose for Rheumatoid arthritis: 90 mg once daily
- Recommended dose for Ankylosing spondylitis: 90 mg once daily
- Recommended dose for Acute gouty arthritis: 120 mg once daily for 8 days
- Recommended dose for Postoperative dental surgery pain: 90 mg once daily, max 3 days
- Shortest duration possible and the lowest effective daily dose should be used
Interaction
- E-coxib may increase prothrombin time when administered with oral anticoagulants
- May reduce the effect of diuretics and other antihypertensive drugs when used with Diuretics, ACE inhibitors, and Angiotensin II Antagonists
- Can be used concomitantly with low-dose acetylsalicylic acid for cardiovascular prophylaxis
- Potential increase in nephrotoxic effects when coadministered with ciclosporin or tacrolimus
- Decreases lithium renal excretion and therefore increases lithium plasma levels when used with Lithium
Contraindications
- Hypersensitivity to the active substance or to any of the excipients
- Active peptic ulceration or active GI bleeding
- Patients with previous bronchospasm, acute rhinitis, nasal polyps, etc. after taking NSAIDs
- Pregnancy and lactation
- Severe hepatic dysfunction, estimated renal creatinine clearance <30 ml/min
- Children and adolescents under 16 years of age
- Inflammatory bowel disease, congestive heart failure, uncontrolled hypertension, established ischemic heart disease, peripheral arterial disease, and/or cerebrovascular disease

Pregnancy & Lactation
- Not recommended in women attempting to conceive
- Excreted in the milk of lactating rats
- Women who use Etoricoxib must not breastfeed
Precautions & Warnings
- Caution advised with treatment of patients at risk of developing GI complication with NSAIDs
- Patients with significant risk factors for cardiovascular events should be treated with caution
- May cause a reduction in prostaglandin formation and impair renal function
- Exercise caution in patients with a history of cardiac failure, left ventricular dysfunction, or hypertension
- Monitor liver function, discontinue at the first appearance of hypersensitivity
Overdose Effects
Administration of single doses up to 500 mg and multiple doses up to 150 mg/day did not result in significant toxicity
