Indications

Pharmacology
- Biguanide type oral antihyperglycemic drug
- Lowers basal and postprandial plasma glucose

- Improves insulin sensitivity

Interaction
- Potential interaction with Furosemide
- Enhancement of absorption by Nifedipine

Contraindications
- Hypersensitivity to the active substance or excipients
- Acute metabolic acidosis
- Severe renal failure
- Acute conditions with potential to alter renal function
- Acute or chronic diseases causing tissue hypoxia
Side Effects
- Hemolytic anemia
- Lactic acidosis
- Decrease of vitamin B12 absorption
- Peripheral neuropathy in patients with vitamin B12 deficiency
- Taste disturbance
- Gastrointestinal disorders (nausea, vomiting, diarrhea)
- Liver function test abnormalities or hepatitis
Pregnancy & Lactation
- Increased risk of congenital abnormalities and perinatal mortality
- Breastfeeding not recommended during treatment
Precautions & Warnings
- Excreted by the kidney
- Potential for lactic acidosis
- Lowering of vitamin B12 levels
- Increased risk of hypoglycemia
- Regular monitoring of renal function advised
- Therapy should be stopped before certain clinical investigations
- Annual estimation of Vitamin B-12 levels recommended
Use in Special Populations
- Adjusted dosage for elderly subjects based on renal function
- No effect on growth and puberty detected in controlled clinical studies
- Regular assessment of renal function necessary

Overdose Effects
- Lactic acidosis is a medical emergency
