Indications
- Treatment of type 2 diabetes mellitus in adults
- Improves glycaemic control
- Can be used as monotherapy
- Can be used in combination therapy
- For patients inadequately controlled by diet and exercise
- For patients for whom metformin is inappropriate
- For patients with renal impairment
Pharmacology
- Linagliptin is an inhibitor of DPP-4 enzyme
- Increases concentrations of active incretin hormones
- Stimulates insulin release from pancreatic beta cells
- Decreases glucagon secretion from pancreatic alpha cells
- Improves glycemic control in type 2 diabetes mellitus
Dosage & Administration
- Linagliptin 5 mg once daily
- Can be taken with or without a meal
- No dose adjustment required for renal impairment
- Maintain dose when added to metformin
- Consider lower dose of sulphonylurea in combination therapy
Interaction
- Weak competitive and mechanism-based inhibitor of CYP3A4
- Low risk for clinically meaningful interactions
- No clinically relevant effects on pharmacokinetics of other medications
Contraindications
- Hypersensitivity to active substance or excipients
