Description
- Atasin is indicated for reducing total cholesterol, LDL cholesterol, apolipoprotein B, and triglycerides.
- It is to be used as an adjunct to diet when other non-pharmacological measures are inadequate.
- Atorvastatin is a selective inhibitor of HMG-CoA reductase, lowering plasma cholesterol and lipoprotein levels.
- Dosage ranges from 10 to 80 mg once daily, adjusted according to individual response.
- Common side effects include constipation, flatulence, dyspepsia, abdominal pain, infection, headache, and myalgia.
- Atorvastatin is contraindicated during pregnancy and lactation, and in patients with active liver disease or unexplained elevations of serum transaminases.
Indications
- Reduction of total cholesterol and LDL cholesterol in heterozygous and homozygous familial hypercholesterolaemia
- Reduction of elevated cholesterol and triglycerides in patients with mixed dyslipidemia
- Treatment of patients with elevated serum triglyceride levels in hypertriglyceridaemia
- Treatment of patients with dysbetalipoproteinaemia
- Reduction of cardiac ischaemic events in patients with coronary artery disease
- Reduction of cholesterol levels associated with diabetes mellitus or renal transplantation
Pharmacology
- Atorvastatin is rapidly absorbed after oral administration with maximum plasma concentrations within 1 to 2 hours
- Mean volume of distribution of Atorvastatin is approximately 381 liters with 98% plasma protein binding
- Extensively metabolized to ortho- and parahydroxylated derivatives
- Atorvastatin and its metabolites are eliminated primarily in bile
Dosage & Administration
- Adults: Usually 10 mg once daily; may be increased to max. 80 mg once daily if necessary
- Children (10-18 years): Initially 10 mg once daily, increased to max. 20 mg once daily if necessary
Interactions
- Risk of myopathy increased with concurrent administration of cyclosporine, fibric acid derivatives, niacin, erythromycin, and azole antifungals
- Plasma concentrations of atorvastatin decreased when coadministered with antacid suspension or increased when coadministered with digoxin, erythromycin, or oral contraceptives
Contraindications
- Hypersensitivity to any component of the medication
- Active liver disease or unexplained persistent elevations of serum transaminases
- History of serious adverse reaction to prior administration of HMG-CoA reductase inhibitors

Pregnancy & Lactation

Precautions & Warnings
- Liver function tests should be performed before the initiation of treatment and periodically thereafter
- Atasin should be used with caution in patients who consume substantial quantities of alcohol or have a history of liver disease
Use in Special Populations
- Plasma concentrations of Atasin are higher in healthy elderly subjects than in young adults
- No clinically significant difference in LDL-C reduction with Atasin between men and women
- Renal disease has no influence on the plasma concentrations or LDL-C reduction of Atasin
- In patients with chronic alcoholic liver disease, plasma concentrations of Atasin are markedly increased
Overdose Effects
- Specific treatment is not available for atorvastatin overdose
- Liver function tests should be performed and serum CK levels should be monitored
