Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
- Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
- Binds with high affinity to the alpha2-delta site in central nervous system tissues
Dosage
- Maximum recommended dose of 100 mg three times a day for adults with DPN
- Recommended dose of 75 to 150 mg two times a day for PHN in adults
- Dosage range of 300 to 450 mg/day for fibromyalgia in adults
- Starting dose range of 150 to 600 mg/day for neuropathic pain associated with spinal cord injury in adults
Administration
- Oral route of administration
- Can be taken with or without food
- Pregabalin CR tablet should be administered after an evening meal
- Instructions on how to take missed doses of Pregabalin CR
Interaction
- Unlikely to be involved in significant pharmacokinetic drug interactions
Contraindications
- Known hypersensitivity to Pregabalin or any of its components

- Weight gain
- Difficulty with concentration/attention
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Small amounts detected in the milk of lactating women
- Breastfeeding not recommended due to potential risk of tumorigenicity
Precautions & Warnings
- Risk of angioedema
- Risk of hypersensitivity reactions
- Increased risk of suicidal thoughts or behavior
- Respiratory depression may occur
- Risk of increased seizure frequency if rapidly discontinued
Use in Special Populations
- Safety and effectiveness not established for pediatric patients
- Pediatric patients below 1 month for partial onset seizures not established
- Safety and effectiveness of Pregan extended-release tablet in pediatric patients not established

- Reduced consciousness
- Depression/anxiety
- Confusional state
- Agitation and restlessness
- Seizures
- Heart block
- No specific antidote
