Indications
- Treatment of hypertension
- Reduction of stroke risk in patients with hypertension and left ventricular hypertrophy
Pharmacology
- Losartan and its active metabolite block vasoconstrictor and aldosterone-secreting effects of angiotensin II
- Hydrochlorothiazide affects renal tubular mechanisms of electrolyte reabsorption
Dosage
- Usual starting dose of 50/12.5 is one tablet once daily
- Maximum dose is 100/25 once daily
- Antihypertensive effect is attained within three weeks after initiation of therapy
Administration
- May be administered with or without food
- May be administered with other antihypertensive agents
Interaction
- No significant drug-drug pharmacokinetic interactions found with certain drugs
- Potential interactions with antidiabetic drugs, other antihypertensive drugs, and anionic exchange resins
Contraindications
- Contraindicated in patients hypersensitive to any component
- Contraindicated in patients with anuria or hypersensitivity to sulfonamide-derived drugs

- Periodic determination of serum electrolytes
- Hypokalemia may rarely develop
- Impaired renal function
- Symptomatic hypotension

Overdose Effects
- Manifestations of overdosage include hypotension and tachycardia
- Supportive treatment should be instituted if symptomatic hypotension occurs
