Indications
- Duodenal ulcer
- Gastric ulcer
- Chronic gastritis
- Prophylaxis of gastrointestinal hemorrhage from stress ulceration in seriously ill patients
Pharmacology
- Non-systemic drug
- Minimally absorbed from the gastrointestinal tract
- Promotes healing of ulcers by forming a protective barrier
- Inhibits pepsin and bile action
Dosage
- Usual dose for adults: 2 gm twice daily or 1 gm four times a day
- Maximum daily dose: 8 gm
- Pediatric population safety and efficacy not established
- No special dosage requirements for elderly patients
Administration
- Should be taken on an empty stomach
- Antacid should not be administered within 30 minutes of Sucralfate
Interaction
- May reduce bioavailability of certain drugs like Fluoroquinolones, Tetracycline, Ketoconazole, etc.
- Separate administration from other agents when critical for bioavailability
- Not to be co-administered with citrate preparations
Contraindications
- Contraindicated in patients with hypersensitivity to sucralfate
Side Effects
- Common adverse events: headache, nausea, abdominal pain, constipation, diarrhea, urticaria
- Bezoars reported in patients with underlying medical conditions or receiving enteral tube feedings
- Episodes of hyperglycemia reported in diabetic patients
Pregnancy & Lactation
- Safety not established in pregnant women
- Caution advised during breastfeeding
Precautions & Warnings
- Use with caution in patients with renal dysfunction
- Not recommended for individuals on dialysis
- Periodic laboratory testing recommended for patients with renal impairment
- Bezoars reported mainly in severely ill patients in ICU
- Dose adjustments not necessary for elderly patients
Use in Special Populations
- Not recommended for use in children under 14 years of age due to insufficient data on safety and efficacy
- Dose adjustments not necessary for elderly patients
- Use with caution in renal insufficiency patients
Overdose Effects
- Most cases of overdose remained asymptomatic
- Symptoms of abdominal pain, nausea, and vomiting reported in few cases
- Minimal risk associated with overdose
