Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
- Structural derivative of inhibitory neurotransmitter gamma-aminobutyric acid (GABA)
- Binds with high affinity to the alpha2-delta site in central nervous system tissues
Dosage
- Maximum recommended dose for diabetic peripheral neuropathy: 100 mg three times a day
- Maximum recommended dose for fibromyalgia: 300 to 450 mg/day
- Dosage form: Pregabalin tablet and Pregabalin CR capsule
- Route of administration: Oral
- Dosage adjustment based on individual patient response and tolerability
Administration
- Pregabalin CR tablet should be administered after an evening meal
- Swallowed whole and should not be split, crushed, or chewed
- Gradual tapering when discontinuing both Pregabalin and Pregabalin CR over a minimum of 1 week
Interaction
- Unlikely to be involved in significant pharmacokinetic drug interactions
- No significant drug interaction with food and others
Contraindications
- Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components
Side Effects
- Common side effects in adults: dizziness, somnolence, dry mouth, edema, blurred vision, weight gain
- Common side effects in pediatric patients: increased weight and increased appetite
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Small amounts detected in the milk of lactating women
Precautions & Warnings
- Angioedema may occur and may be associated with life-threatening respiratory compromise
- Patients need to be monitored when used with concomitant CNS depressants
- Pregabalin may cause dizziness and somnolence impairing patients' ability to drive or operate machinery
- Increased seizure frequency or adverse reactions may occur if Pregabalin is rapidly discontinued
Use in Special Populations
- Safety and effectiveness in pediatric patients not established for certain indications
- Safety and effectiveness not established in pediatric patients below the age of 1 month
Overdose Effects
- Signs and symptoms: reduced consciousness, depression/anxiety, seizures
- No specific antidote
- General supportive care indicated including monitoring of vital signs
