Description
- Selective and competitive inhibitor of HMG-CoA reductase
- Indicated for Heterozygous Hypercholesterolemia, Homozygous Hypercholesterolemia, Mixed Dyslipidemia, Primary prevention of cardiovascular disease
Indications
- Heterozygous Hypercholesterolemia (Familial and Non familial)
- Homozygous Hypercholesterolemia (Familial)
- Mixed Dyslipidemia (Fredrickson Type IIa and IIb)
- Primary prevention of cardiovascular disease
Pharmacology
- Selective and competitive inhibitor of HMG-CoA reductase
- Increases hepatic LDL receptors
- Inhibits hepatic synthesis of VLDL and LDL particles
Dosage
- Dose range: 5-40 mg once daily
- Starting dose: 20 mg/day for HoFH
- Pediatric patients: 5-10 mg/day for 8 to less than 10 years age, 5-20 mg/day for 10 to 17 years of age
Administration
- Can be taken with or without food, at any time of day
Interaction
- Cyclosporine: Combination increases exposure, dose limited to 5 mg once daily
- Gemfibrosil: Combination should be avoided, if used together dose limited to 10 mg once daily
- Lopinavir/Ritonavir or atazanavir/ritonavir: Combination increases exposure, dose limited to 10 mg once daily
- Coumarin anticoagulants: Combination prolongs international normalized ratio (INR), requires frequent monitoring
Contraindications
- Known hypersensitivity to product components
- Liver disease
- Pregnant women and women who may become pregnant
- Nursing mothers

- Constipation
- Asthenia
- Abdominal pain
- Nausea
Precautions warnings
- Skeletal muscle effects (e.g., myopathy and rhabdomyolysis)
- Liver enzyme abnormalities and monitoring
- Use in children not established
