Dosage range
- 5-40 mg once daily
Recommended starting dose
- 20 mg/day for HoFH
- 5-10 mg/day for pediatric patients aged 8 to less than 10 years
- 5-20 mg/day for pediatric patients aged 10 to 17 years
- 20 mg/day for pediatric patients aged 7 to 17 years
Administration
- Can be taken with or without food, at any time of day
Pharmacology
- Inhibits HMG-CoA reductase enzyme
- Increases hepatic LDL receptor count
- Inhibits hepatic synthesis of VLDL and LDL particles
Indications
- Heterozygous Hypercholesterolemia (Familial and Non-familial)
- Homozygous Hypercholesterolemia (Familial)
- Mixed Dyslipidemia (Fredrickson Type IIa and IIb)
- Primary prevention of cardiovascular disease
Drug interactions
- Cyclosporine
- Gemfibrosil
- Lopinavir/Ritonavir or atazanavir/ritonavir
- Coumarin anticoagulants
- Concomitant lipid-lowering therapies
Contraindications
- Known hypersensitivity to product components
- Liver disease with unexplained persistent elevations in hepatic transaminase levels
- Pregnant women and women who may become pregnant
- Nursing mothers
