Indications
- Treatment of secondary amenorrhea
- Treatment of abnormal uterine bleeding
- Reduction of endometrial hyperplasia
- Indicated for nonhysterectomized postmenopausal women
Pharmacology
- Transforms proliferative into secretory endometrium
- Androgenic and anabolic effects
- Devoid of significant estrogenic activity
- Inhibits gonadotropin production
Dosage & Administration
- Dosage of 5 or 10 mg daily for 5 to 10 days
- Daily dose of 10 mg for 10 days for optimum secretory transformation
- Dosage beginning on the 16th or 21st day of the menstrual cycle
- Reduction of endometrial hyperplasia dosage
Contraindications
- Undiagnosed abnormal genital bleeding
- Known or suspected breast cancer
- Estrogen- or progesterone-dependent neoplasia
- Active DVT, PE, or history of these conditions
Side Effects
- Abnormal uterine bleeding
- Breast tenderness
- Thromboembolic disorders
- Nausea and cholestatic jaundice
Pregnancy & Lactation
- Should not be used during pregnancy
- Increased risks for hypospadias, clitoral enlargement and labial fusion
- Not to be used during lactation
Precautions & Warnings
- Lowered incidence of endometrial hyperplasia
- Monitoring of blood pressure
- Hypertriglyceridemia precautions
- Hepatic impairment and cholestatic jaundice precautions
Use in Special Populations
- Not indicated in children
- Insufficient studies conducted in the pediatric population
- Effectiveness in geriatric women
