Indications
- Heterozygous Hypercholesterolemia (Familial and Non familial)
- Homozygous Hypercholesterolemia (Familial)
- Mixed Dyslipidemia (Fredrickson Type IIa and IIb)
- Primary prevention of cardiovascular disease
Pharmacology
- Selective and competitive inhibitor of HMG-CoA reductase
- Increases hepatic LDL receptors
- Inhibits hepatic synthesis of VLDL
Dosage
- Dose range: 5-40 mg once daily
- Starting dose for HoFH: 20 mg/day
- Pediatric patients with HeFH: 5-20 mg/day
- Pediatric patients with HoFH: 20 mg/day
Administration
Can be taken with or without food, at any time of day
Interaction
- Cyclosporine: Limit Rosu dose to 5 mg once daily
- Gemfibrosil: Avoid combination or limit Rosu dose to 10 mg once daily
- Lopinavir/Ritonavir or atazanavir/ritonavir: Limit Rosu dose to 10 mg once daily
- Coumarin anticoagulants: Prolongs INR, monitor frequently
- Concomitant lipid-lowering therapies: Risk of skeletal muscle effects
Contraindications
- Known hypersensitivity to product components
- Liver disease
- Pregnant women and women who may become pregnant
- Nursing mothers
