Description
- Non-systemic medication for adult and adolescent ulcer treatment
- Promotes healing of gastric and duodenal ulcers
- Contraindicated in patients with hypersensitivity to sucralfate
Dosage
- Usual dose for duodenal ulcer and gastric ulcer in adults is 2 gm of sucralfate twice daily
- Usual dose for prophylaxis of gastrointestinal hemorrhage from stress ulceration in adults is 1 gm of sucralfate 4-6 times a day
- No established safety and efficacy data for children under 14 years of age
- No special dosage requirements for elderly patients
Administration
- Should be taken on an empty stomach
- Antacids should not be administered within 30 minutes of sucralfate
Interaction
- May reduce bioavailability of fluoroquinolones, tetracycline, ketoconazole, etc.
- Separate administration from certain drugs may restore bioavailability
- Should not be co-administered with citrate preparations
Pharmacology
- Non-systemic as the drug is minimally absorbed from the gastrointestinal tract
- Promotes healing by forming a chemical complex that binds to the ulcer site
- Inhibits the action of pepsin and bile
Indications
- Treatment of duodenal ulcer, gastric ulcer, chronic gastritis
- Prophylaxis of gastrointestinal hemorrhage from stress ulceration in seriously ill patients
Contraindications
- Contraindicated in patients with hypersensitivity to sucralfate
Side Effects
- Common adverse events include headache, nausea, abdominal pain, constipation, diarrhea, and urticaria
- Episodes of hyperglycemia reported in diabetic patients
- Bezoar formation reported in some patients
Pregnancy & Lactation
- Safety in pregnant women not established, use only if clearly needed
- Caution in breastfeeding women
Precautions & Warnings
- Use with caution in patients with renal dysfunction
- Not recommended for individuals on dialysis
Use in Special Populations
- Not recommended for use in children under 14 years of age
- No dose adjustments necessary in elderly patients
Overdose Effects
- Most cases of overdose remained asymptomatic
- Symptoms of abdominal pain, nausea, and vomiting reported in a few cases
- Acute oral toxicity studies in animals found minimal risks associated with overdose
