Indications
- Heterozygous Hypercholesterolemia (Familial and Non familial)
- Homozygous Hypercholesterolemia (Familial)
- Mixed Dyslipidemia (Fredrickson Type IIa and IIb)
- Primary prevention of cardiovascular disease
Pharmacology
- Selective and competitive inhibitor of HMG-CoA reductase
- Increases the number of hepatic LDL receptors
- Inhibits hepatic synthesis of VLDL
Dosage
- Dose range: 5-40 mg once daily
- Starting dose for HoFH: 20 mg/day
- Pediatric patients with HeFH: 5-10 mg/day for 8 to less than 10 years age, 5-20 mg/day for 10 to 17 years of age
- Pediatric patients with HoFH: 20 mg/day for 7 to 17 years of age
Administration
- Can be taken with or without food, at any time of day
Interaction
- Remarkable drug interactions with Cyclosporine, Gemfibrosil, Lopinavir/Ritonavir, atazanavir/ritonavir, Coumarin anticoagulants, Concomitant lipid-lowering therapies
Contraindications
- Known hypersensitivity to product components
- Liver disease
- Pregnant women and women who may become pregnant
- Nursing mothers

- Constipation
- Asthenia
- Abdominal pain
- Nausea
Pregnancy & Lactation
- Safety in pregnant women has not been established
- Excretion in human milk not known
Precautions & Warnings
- Skeletal muscle effects increase risk with 40 mg dose, advanced age, hypothyroidism, renal impairment
- Prompt reporting of unexplained muscle pain, tenderness or weakness
- Monitoring of liver enzymes before and during treatment
