Indications
- Treatment of type 2 diabetes mellitus in adults
- Monotherapy in inadequately controlled patients
- Combination therapy with metformin
- Combination therapy with sulphonylurea and metformin
Pharmacology
- Inhibitor of DPP-4 enzyme
- Increases concentrations of active incretin hormones
- Stimulates insulin release in a glucose-dependent manner
- Decreases glucagon secretion
Dosage & Administration
- 5 mg once daily
- Concomitant administration with metformin
- Consideration for lower dose of sulphonylurea in combination therapy
- No dose adjustment required for renal impairment
- Can be taken with or without a meal at any time of the day
Interaction
- Weak competitive and weak to moderate mechanism-based inhibitor of CYP3A4
- Low risk for clinically meaningful interactions with other medicinal products
- Had no clinically relevant effect on the pharmacokinetics of metformin, glyburide, simvastatin, warfarin, digoxin, or oral contraceptives
