Indications
- Kinase inhibitor
- Treatment of HR-positive, HER2-negative advanced breast cancer
- Combination therapy with aromatase inhibitor or fulvestrant
Pharmacology
- Inhibitor of cyclin-dependent kinase (CDK) 4 and 6
- Regulates cell cycle progression
- Reduced cell proliferation in breast cancer cell lines
Dosage & Administration
- Oral administration
- Taken with or without food
- Recommended starting dose: 600 mg orally once daily for 21 days
Interaction
- Avoid concomitant use with strong CYP3A inhibitors
- Dose adjustment required with sensitive CYP3A substrates
- Avoid concomitant use with drugs known to prolong QT interval
Side Effects
- Common adverse reactions including leukocytes decreased, infections, nausea, fatigue, diarrhea
- Laboratory abnormalities such as hemoglobin decreased, platelets decreased, glucose serum decreased
Pregnancy & Lactation
- Can cause fetal harm
- No data on presence in human milk or effects on breastfed infant
Precautions & Warnings
- Monitor for pulmonary symptoms indicative of ILD/pneumonitis
- Permanently discontinue in case of severe ILD/pneumonitis or severe cutaneous adverse reactions
- Monitor ECGs and electrolytes for QT interval prolongation
- Avoid concomitant use with tamoxifen
- Monitor liver function tests for hepatobiliary toxicity
- Monitor CBC for neutropenia
- Advice females of reproductive potential to use effective contraception during therapy
