Indications

  • Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
  • Postherpetic neuralgia (PHN)
  • Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
  • Fibromyalgia
  • Neuropathic pain associated with spinal cord injury

Mode of Action

Structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA), binds with high affinity to the alpha2-delta site in central nervous system tissues

Dosage

  • DPN Adult:
    • 50-100 mg three times a day, maximum 300 mg/day
    • Begin CR capsule at 165 mg once daily, increase to 330 mg once daily
  • PHN Adult:
    • 75-150 mg two times a day or 50-100 mg three times a day, maximum 300 mg/day
    • Begin CR capsule at 165 mg once daily, increase to 330 mg once daily
  • Fibromyalgia:
    • 300-450 mg/day
    • Begin at 75 mg two times a day, increase to 150 mg two times a day
  • Spinal Cord Injury:
    • 150-600 mg/day
    • Start at 75 mg two times a day, increase to 150 mg two times a day
  • Conversion From Pregabalin Capsules to Pregabalin CR Capsule Tablet: Dosing equivalence provided

Administration

  • Route: Oral, with or without food
  • CR Tablet: Administered after an evening meal, swallow whole, not split, crushed, or chewed

Interaction

  • Medication: Unlikely to have significant pharmacokinetic drug interactions
  • Food and Others: Not applicable

Contraindications

Hypersensitivity to Pregabalin or any of its components

    • Dry mouth
    • Edema
    • Blurred vision
    • Weight gain
    • Difficulty with concentration/attention
  • Pediatric Patients:
    • Increased weight
    • Increased appetite

Pregnancy & Lactation

No adequate and well-controlled studies, small amounts detected in the milk of lactating women, breastfeeding not recommended
