Indications

Dihydropyridine calcium antagonist
Inhibits transmembrane influx of calcium ions into vascular smooth muscle and cardiac muscle
Selective effect on vascular smooth muscle cells
Greater effect on vascular smooth muscle cells than on cardiac muscle cells
1:1 racemic mixture of levamlodipine and dextro amlodipine
Pharmacologically active isomer is levamlodipine
Oral administration
Takes 6-12 hours to reach peak concentration (Tmax)
Bioavailability of 64-90%
Volume of distribution similar to amlodipine (21 L/kg)
Metabolized to inactive metabolites
Metabolism primarily mediated by CYP3A4
60% eliminated in urine
10% eliminated as unmetabolized drug

CYP3A inhibitors increase systemic exposure to amlodipine
CYP3A inducers effects not quantitatively available
Co-administration with simvastatin increases the systemic exposure of simvastatin
May increase systemic exposure of cyclosporine or tacrolimus when co-administered
Can be taken with or without food
Avoid grapefruit products and natural licorice

Common: Difficulty in breathing, Dizziness, Reflex Tachycardia, Hypotension, Abdominal Pain, Edema
Rare: Diarrhea, Back pain, Headache, Fatigue, Nausea
