Indications

Pharmacology
- Dihydropyridine calcium antagonist
- Inhibition of calcium ion influx
- Selective effect on vascular smooth muscle cells
- Volume of distribution similar to amlodipine
- Metabolized by CYP3A4
- Excretion in urine
Dosage & Administration
- Oral route of administration
- Usual initial dose of 2.5 mg once daily
- Maximum dose of 5 mg once daily
- Reduced dose for elderly and hepatic insufficiency
- Titration for dosage adjustment
- Pediatric dose of 1.25 mg to 2.5 mg once daily
Interaction
- Increased systemic exposure with CYP3A inhibitors
- No quantitative effects with CYP3A inducers
- Increased systemic exposure of simvastatin
- Increased systemic exposure of immunosuppressants
- Can be taken with or without food
- Avoid grapefruit products and natural licorice
Contraindications
- Known sensitivity to amlodipine

- Slow titration for patients with hepatic impairment

- Excessive peripheral vasodilation
- Marked hypotension
- Possibly reflex tachycardia
