Pharmacology
- Potentiation of serotonergic activity
- Inhibition of neuronal reuptake of serotonin
- Blockage of serotonin uptake into human platelets
- Selective inhibitor of neuronal serotonin reuptake
- Little affinity for muscarinic, alpha1, alpha2, beta adrenergic, dopamine (D2), 5-HT1, 5 HT2, and histamine (H1)-receptors
Dosage & Administration
- Usual initial dosage of 20 mg/day for Major Depressive Disorder
- Maintenance therapy at doses averaging about 30 mg
- Usual initial dosage of 40 mg/day for Obsessive Compulsive Disorder
- Maintenance therapy with periodic reassessment
- Usual initial dosage of 40 mg/day for Panic Disorder
- Maximum dosage of 60 mg/day
- Usual initial dosage of 20 mg/day for Social Anxiety Disorder
- Usual initial dosage of 20 mg/day for Generalized Anxiety Disorder
- Usual initial dosage of 20 mg/day for Posttraumatic Stress Disorder
Interaction
- Absorption and pharmacokinetics not affected by food or antacids
- Potential interaction between Oxat and Tryptophan
- Effects on drug metabolizing enzyme inducers/inhibitors
- Advice against concomitant use of Oxat and alcohol in depressed patients
- Limited effect on sedation and drowsiness when combined with certain drugs
- Potential interaction with MAO inhibitors, lithium, phenytoin/anticonvulsants, and warfarin
Contraindications
- Concomitant use with MAOIs or thioridazine
- Hypersensitivity to Paroxetine
