Indications
- Treatment of chorea associated with Huntington's disease
Pharmacology
- Tetrabenazine as a vesicular monoamine transporter-2 (VMAT-2) inhibitor
Dosage & Administration
- Can be taken with or without food
- Individualization of dose with careful weekly titration is required
- Starting dose: 12.5 mg daily
- Maximum daily dose: 100 mg for extended and intermediate metabolizers
- Pediatric & geriatric use not established
Interaction
- Strong CYP2D6 Inhibitors may require reduction in Ticstop dose
- At least 20 days should elapse after stopping reserpine before starting Ticstop
- No interaction with food
Contraindications
- Actively suicidal, untreated or undertreated Hepatic impairment
- Concurrent use of monoamine oxidase inhibitors (MAOIs) or reserpine
- Concurrent use of deutetrabenazine or valbenazine
Side Effects
- Common adverse reactions: Sedation/somnolence, fatigue, insomnia, depression, akathisia, anxiety/anxiety aggravated & nausea
- Rare adverse reactions: Suicidal thoughts, neuroleptic malignant syndrome, pneumonia
Pregnancy & Lactation
- No adequate data on developmental risk during pregnancy
- No data on presence in human milk
Precautions & Warnings
- Benefit and potential adverse effects should be periodically re-evaluated
- Maximum daily dose not to exceed 50 mg/day
- Treatment should be discontinued in case of Neuroleptic Malignant Syndrome
- May impair patient's ability to drive or operate machinery
- Not recommended in combination with other drugs that prolong QTc
Overdose Effects
- Signs of overdosage: drowsiness, sweating, hypothermia
