Sinapride 2 mg (Tablet)

Indications

• Symptomatic treatment of chronic constipation in adults

Pharmacology

• Selective stimulator of 5-HT4 receptors
• Stimulates motility by interacting specifically with 5-HT4 receptors
• Causes release of acetylcholine and further contraction of muscle layer of the colon
• Relaxes circular muscle layer leading to the propulsion of luminal content

Dosage & Administration

• 2 mg once daily for adults
• 1 mg once daily for older people
• Not recommended for children and adolescents younger than 18 years

Interaction

• Low interaction potential
• Not expected to affect the CYP-mediated metabolism of co-medicated medicinal products
• Not an inhibitor of P-gp at clinically relevant concentrations
• Potential increase in systemic exposure with potent inhibitors of P-gp

Contraindications

• Hypersensitivity to the active substance or excipients
• Renal impairment requiring dialysis

Side Effects

• Frequently reported adverse reactions: headache, abdominal pain, nausea, diarrhea
• Majority of adverse events were mild to moderate in intensity

Pregnancy & Lactation

• Not recommended during pregnancy
• Effective contraception recommended during treatment
• Not recommended during breastfeeding

Precautions & Warnings

• Dose adjustment for severe renal impairment
• Limited data in patients with severe hepatic impairment
• Possible reduction in efficacy of oral contraceptives in case of severe diarrhea
• Contains lactose: Not suitable for patients with rare hereditary problems of galactose intolerance, Lapp lactase deficiency or glucose-galactose malabsorption

Use in Special Populations

• Dose adjustment for patients with severe renal impairment
• Dose adjustment for patients with severe hepatic impairment
• No dose adjustment required for patients with mild to moderate renal or hepatic impairment

Overdose Effects

• Symptoms may include headache, nausea, and diarrhea
• Specific treatment not available
• Supportive measures instituted as required
• Correction of electrolyte disturbances may be necessary

• Store at room temperature, below 30°C
• Do not remove desiccant
• Dispense in original bottle
