Indications
- Neuropathic pain associated with diabetic peripheral neuropathy (DPN)
- Postherpetic neuralgia (PHN)
- Adjunctive therapy for the treatment of partial-onset seizures in patients 1 month of age and older
- Fibromyalgia
- Neuropathic pain associated with spinal cord injury
Pharmacology
Pregabalin is a structural derivative of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA). It binds with high affinity to the alpha2-delta site in central nervous system tissues, resulting in anti-nociceptive and antiseizure effects.
Dosage
- Maximum recommended dose for diabetic peripheral neuropathy (DPN): 300 mg/day
- Maximum recommended dose for postherpetic neuralgia (PHN): 300 mg/day
- Recommended dose for fibromyalgia: 300 to 450 mg/day
- Recommended dose for spinal cord injury: 150 to 600 mg/day
Administration
- Route of administration: Oral
- Pregabalin CR tablet administration: After an evening meal, swallowed whole and should not be split, crushed or chewed
Interaction
- Drug interaction with medication: Unlikely to be involved in significant pharmacokinetic drug interactions
Contraindications
Contraindicated in patients with known hypersensitivity to Pregabalin or any of its components
Side Effects
- Most common side effects in adults:dizziness, somnolence, dry mouth, edema, blurred vision, weight gain, thinking abnormal
- Most common side effects in pediatric patients:increased weight, increased appetite
Pregnancy & Lactation
Not recommended during treatment due to potential risk to the fetus. Small amounts of pregabalin detected in the milk of lactating women.
Precautions & Warnings
- Angioedema can occur and may be associated with life-threatening respiratory compromise
- Discontinue immediately in case of hypersensitivity reactions
- Increase in the risk of suicidal thoughts or behavior
Use in Special Populations
Safety and effectiveness in pediatric patients have not been established
Overdose Effects
Signs and symptoms include reduced consciousness, depression/anxiety, confusional state, agitation, restlessness, seizures, and heart block
