Description

  • Tablets for the treatment of nausea and vomiting of pregnancy in women who do not respond to conservative management

Pharmacology

  • Antihistamine
  • Histamine (H1) receptor blocker
  • High affinity for H1 receptors in the brain
  • Inhibition of vestibular system & Muscarinic receptor
  • Antiemetic properties of Pyridoxine Hydrochloride

Dosage administration

Oral administration, initially one tablet at bedtime, maximum daily dose of two tablets, on an empty stomach with water, not to be crushed, chewed, or split

Interaction

  • Contraindicated with monoamine oxidase inhibitors (MAOIs)
  • Not recommended with alcohol and other CNS depressants

Contraindications

  • Hypersensitivity to components
  • Monoamine oxidase (MAO) inhibitors use

Side effects

Somnolence, accidents from combined use with CNS depressants

Pregnancy lactation

  • Pregnancy category A
  • Excretion into breast milk, caution during breastfeeding

Precautions warnings

  • May cause somnolence
  • Avoid driving or operating heavy machinery
  • Not recommended with concurrent use of CNS depressants or alcohol
  • Use with caution in women with asthma, increased intraocular pressure, narrow angle glaucoma, stenosing peptic ulcer, pyloroduodenal obstruction, or urinary bladder-neck obstruction

Overdose effects

  • Possible delayed signs and symptoms of intoxication due to extended-release formulation
  • Symptoms include restlessness, dryness of mouth, dilated pupils, sleepiness, vertigo, mental confusion, tachycardia, seizures, rhabdomyolysis, acute renal failure, and death
  • Treatment includes gastric lavage or activated charcoal, whole bowel irrigation, and symptomatic treatment
