Laxana 24 mg (Capsule)

Indications

• Irritable Bowel Syndrome with Constipation (IBS-C)
• Chronic Idiopathic Constipation (CIC) in adults
• Opioid-Induced Constipation (OIC) in adult patients with chronic, non-cancer pain

Pharmacology

• Locally-acting chloride-channel activator
• Increases intestinal secretion
• Increases intestinal motility
• Aids passage of stool
• Relieves symptoms associated with chronic idiopathic constipation
• Activates ClC-2 chloride channels
• Causes efflux of chloride ions into the lumen
• Leads to efflux of sodium ions through a paracellular pathway
• Stimulates recovery of muscosal barrier function

Dosage & Administration

• Recommended dose for IBS-C: 8 mcg twice daily orally with food and water
• Recommended dose for Chronic Idiopathic Constipation and OIC: 24 mcg twice daily orally with food and water
• Dosage adjustment for hepatic impairment

Interaction

• Possible decrease in efficacy when used with diphenylheptane opioids

Contraindications

• Contraindicated in patients with known or suspected mechanical gastrointestinal obstruction

• Full or bloated feeling or pressure in the stomach
• Nausea
• Stomach pain
• Swelling of abdominal or stomach area
• Dyspnea

Pregnancy & Lactation

• Pregnancy category C
• Unknown excretion in human milk
• Caution in nursing women

Precautions & Warnings

• Nausea
• Diarrhea
• Bowel Obstruction
• Concomitant administration of food with Laxana to reduce nausea symptoms
• Thorough evaluation for obstruction in patients with symptoms suggestive of mechanical gastrointestinal obstruction

Overdose Effects

• Adverse reactions due to overdose: nausea, diarrhea, vomiting, dizziness, headache, abdominal pain, flushing/hot flash, retching, dyspnea, pallor, stomach discomfort, anorexia, asthenia, chest discomfort, dry mouth, hyperhidrosis, syncope
