Indications

- 133.333 mg (15%w/w) of Esomeprazole enteric coated pellets equivalent to 20 mg Esomeprazole
- 266.67 mg (15% w/w) of Esomeprazole enteric coated pellets equivalent to 40 mg Esomeprazole

Pharmacology
Inhibits gastric acid by blocking the hydrogen-potassium adenosine triphosphatase enzyme system
Dosage
- Erosive esophagitis: 40 mg once daily for adults (≥18 years), 40 mg once daily for children & adolescents (12-18 years)
- Maintenance of healing of erosive esophagitis: 20 mg once daily for adults (≥18 years), 20 mg once daily for children & adolescents (12-18 years)
- Risk reduction in NSAID associated gastric ulcer: 20 mg once daily for adults (≥18 years) for 4-8 weeks
- H. pylori eradication: 20 mg twice daily for adults (≥18 years) for 7 days, 20 mg twice daily for children & adolescents (12-18 years) for 7 days
- Zollinger-Ellison syndrome and idiopathic hypersecretion: 40-80 mg twice daily for adults (≥18 years)
Administration
Tablets should be swallowed whole with liquid, not chewed or crushed. Can also be dispersed in half a glass of non-carbonated water and should be taken immediately or within 30 minutes. Rinse the glass with half a glass of water and drink. Pellets must not be chewed or crushed.
Interaction
Metabolized in the liver by CYP2C19 and CYP3A4. Not likely to inhibit CYPs 1A2, 2A6, 2C9, 2D6, 2E1 and 3A4. No clinically relevant interactions with drugs metabolized by these CYP enzymes are expected. Potential interference with CYP2C19. No clinically significant interactions with phenytoin, warfarin, quinidine, clarithromycin or amoxicillin. May interfere with the absorption of drugs where gastric pH is important. Co-administration with clarithromycin has led to increased plasma levels of REMMO and 14-hydroxyclarithromycin.
Contraindications
Contraindicated in patients with known hypersensitivity to any of the formulation

Precautions & Warnings
- Use carefully with severe liver dysfunction and severe renal impairment
- May slightly increase the risk of hip, wrist and spine fracture, particularly when taken over a period of more than one year
