Description
- Synthetically prepared Syntocin is indicated for labor induction, stimulation of uterine contractions, prevention or treatment of secondary hemorrhage, adherent placenta, and incomplete abortion. It can also be used to stimulate lactation when advised by a registered Gynaecologist/Physician.
Active Substance
- Synthetically prepared Syntocin identical to the natural occurring hormone from the posterior pituitary gland
Indications
- Induction of labor

Pharmacology
- Syntocin causes contractions of the uterus by increasing intracellular Ca^2+ and activating myosin's light chain kinase. It also stimulates the smooth muscle associated with the secretory epithelium of the lactating breast.
Dosage & Administration
- Intravenous infusion under adequate control
- Dosage can be doubled every 30-45 minutes until the uterus threshold has been attained
- Intramuscular administration should not exceed 0.5-1 IU per injection
- Dosage for prevention or treatment of secondary hemorrhage and adherent placenta
- Dosage for lactation stimulation
- Dosage for incomplete abortion treatment
Interaction
- Severe hypertension when given following prophylactic administration of a vasoconstrictor in conjunction with caudal-block anesthesia
- Modification of cardiovascular effects with cyclopropane anesthesia
Contraindications
- Cephalo-pelvic disproportion
- Abnormal fetal position or skull position
- Overdistention of the uterus
- Severe toxaemia
- Hypertonic uterine dysfunction
- Foetal distress
- Cord presentation and prolapse
- Total placenta previa
- Vasa praevia
Side Effects
- Uterine hypertonicity
- Spasm
- Titantic contraction or rupture of the uterus
- Possibility of increased blood loss and afbrinigenemia
- Severe water intoxication with convulsions and coma
- Nausea
- Vomiting
- Hemorrhage
- Cardiac arrhythmias
- Anaphylactic reaction
- Increased incidence of icterus neonatorum
Pregnancy & Lactation
- Pregnancy category C
- Not known whether Oxytocin is excreted in human milk
Precautions & Warnings
- Uterine sensitivity to Syntocin may vary widely
- Utilize the smallest effective dose to initiate labor
- Use only with adequate medical supervision
- Monitor fetal heart rate and maternal blood pressure
- Use with care if scars of the uterus and cervix uteri exist
Overdose Effects
- Violent uterine contractions leading to fetal bradycardia and arrhythmias
- Excessive laceration of soft tissues
- Uterine rupture
- Possible fetal or maternal death

- Potency maintained for at least 24 hours when diluted with specific infusion fluids; possible direct injection into infusion line with certain fluids
